So excited to work with @JunQiLab on this important study!
New research published in @JExpMed from @DanaFarber’s @JunQiLab and collaborators reveals that AKT inhibitors synergize with EP300 degraders to suppress osteosarcoma growth in vitro and in vivo. These agents positively combine across cancer cells in a manner dependent on control of protein synthesis.

Researchers identified a potential therapeutic vulnerability in a subset of osteosarcoma. In preclinical models, combining EP300 degradation with AKT inhibition slowed tumor growth and prolonged survival. The findings suggest a potential path toward more personalized treatment. bit.ly/3U3DwDt

TLDR: a genetically-defined group of osteosarcoma is sensitive to the EP300 PROTAC JQAD1, due to a balance between degradation and resynthesis by the ribosome. Read on for deets...
This work sets the stage for future work testing more drug-like EP300 degraders with AKT inhibitors in osteosarcoma and indeed, across many, many other types of cancer.

How “specific” are the inhibitors we use? In @nchembio, Sterling and Holtsmark et al reveal a dual inhibitory role for the clinical agent zavondemstat! Findings with implications for target and patient selection in trials, discussed by Drs Ali Nezhady and McKay-Corkum.
