Finding 1: LKB1 loss suppresses oestrogen, progesterone, and androgen receptor signalling.
LKB1 is not just a metabolic kinase, it actively supports hormone-responsive transcriptional programs. Losing it reshapes the hormone landscape of the tumour cell.

Overall, LKB1 deficiency rewires breast cancer in a subtype- and cell-type-specific way, thus reshaping hormone, immune, and metabolic programs.

Finding 3: STK11 mutations in TNBC cluster in the kinase domain and associate with immune evasion.
Finding 4: Mammary Lkb1 deletion alone drives DCIS → invasive carcinoma with 81% penetrance, + mTOR↑, HIF1α↑, Cyclin D1↑, ER↓.
Causality confirmed in vivo. 🐭

New pre-print from the Marignanilab.com!
We profiled LKB1/STK11 loss across breast cancer using 4 complementary approaches and found it does far more than flip the AMPK–mTOR switch.
