Almost 15 years after our initial observation that IRE1 is specifically active in cDC1s, we finally start understanding why… Check out the new study from @victor.bosteels, published in @natcomms.nature.com Paper in press @ rdcu.be/fur4Q

In contrast to most secretory cell types where IRE1 gets activated by accumulation of unfolded proteins in the ER, IRE1 gets activated by cholesterol influx during apoptotic cell engulfment in cDC1s. IRE1-dependent RIDD drives cholesterol efflux during homeostatic DC maturation.

This process is specific to cDC1s and is only required during homeostatic not immunogenic maturation. Thanks to @ERC_Research for supporting this work through the ERC-CoG DCRIDDLE. Thanks to @ugent_fge and @InflamResCen for their fantastic support to my team.

Thrilled to present our unpublished work on how dendritic cells decode LNP-embedded antigenic cargo at the @ESGCT meeting in Rome (bit overwhelmed by the amount of people though😱). For all DC and LNP lovers, come find me at 15h3O in Parallel session B.

Very happy that our review in Nat Rev Imm is out! Trying to shed some light on the many knowns and even more unknowns of homeostatic dendritic cell maturation with @Victor_Bosteels @InflamResCen

Interested in dendritic cell biology and/or IRE1 signaling? Ready for a new challenge in the city of Ghent? If the answer is 2x yes, please check the VIB website for open positions for a PhD student jobso.id/har5 and a postdoc jobso.id/har8 in the @JanssensLab

AGR2 is a pretty interesting protein, especially upon mutation of H117 to Y: this causes massive inflammatory phenotypes in patients. Turns out that the H117Y mutant is also completely incapable of blocking IRE1β activity!

So now we finally know why IRE1β behaves as it does: it just needs its partner in crime, AGR2, to calm down.

Very happy to share our preprint on the discovery on the mucus chaperone AGR2 as a regulator of the goblet cell stress sensor IRE1b! Fantastic job done by @ClootsEva in collaboration with the @VIBEyckermanS Lab. Stay tuned for the peer-reviewed paper!

The ever expanding universe of TAOK3…Very proud that this joint study with @bart_lambrecht got published. Big congratulations to @maes_bastiaan and Farzaneh Fayazpour for a massive amount of work!
New in @JExpMed: @maes_bastiaan, @JanssensLab, @bart_lambrecht et al. @ugent reveal an unexpected role of TAOK3 in regulating ST2+ regulatory T cells in mouse adipose tissue. Absence of TAOK3 sustains Tregs in #obesity and improves metabolic dysfunction. bit.ly/3XjLTI0

To our surprise, we could not confirm a role for IRE1 in regulating dendritic cell-mediated anti-tumor immunity, and this despite the use of several tumor models, several animal houses and several Cre-lines to excise the gene from DCs.
Happy to share our collaborative work with @JanssensLab!. Long story in the making but finally is out and our groups are proud: Nuanced role for dendritic cell intrinsic IRE1 RNase in the regulation of antitumor adaptive immunity. doi.org/10.3389/fimmu.2023.1…

It is always difficult to get negative data published, even though they are solid. Still, we were convinced it was important to convey this message to the scientific community in view of the broad role of the UPR in anti-tumor immunity.
