Our crisprVerse paper is out in @NatureComms.
We developed a one-stop shop CRISPR gRNA design tool that works with any nuclease and genome, and includes functionalities for CRISPRko, CRISPRa, CRISPRi, base editing, and RNA-editing.

and then show how our modular approach can easily create rich annotations for different CRISPR modalities using public data. This includes base editing

Finally we show that genetic variation, eg SNP, can influence knockout efficiency as a result of introducing variation in the sgRNA:gDNA binding.
For instance a subset of melanoma cell lines (expressing SOX10, but not expressing SOX9) appear to be sensitive to SOX9 knockout because of off-targets to SOX10
Then we show that off-targets caused by single-mismatch and double-mismatch in the sgRNA:gDNA binding can lead to an incorrect estimation of cancer vulnerabilities when not taken into account
We first show that multi-target guides (guides with several on-targets, for instance paralog genes) are hard to model and should be interpreted with caution because of possible genetic interactions between the on-targets.
