Multiplexing with primary antibodies from the same species has always been a challenge. The Premix&Stain protocol with Smart Secondary Nanobodies addresses this directly. And here is a 5-color example showing what that looks like in practice.
Three targets were labeled using mouse primary antibodies, detected with Smart Secondaries (FluoTag-X2 anti-mouse) plus direct GFP tag labeling for a fourth channel via FluoTag-X2 anti-GFP. Phalloidin STAR BLUE serves as a structural counterstain for actin.

High mtDNA copy number promotes lung cancer growth in the mouse, whereas mtDNA depletion in tumor cells reduces tumor growth. This tumor-intrinsic role for mtDNA can be exploited for development of future lung cancer therapies.
