This capacity is particularly interesting for microbiome editing strategies or to program engineered probiotics to fight antibiotic-resistant infections as recently described by our group.

Each PilV variant generated by the shufflon acts as a different adhesin to recognize specific groups of bacteria and allow conjugation. The shufflon is thus a molecular switch expanding and tuning the ability of TP114 to disseminate in the microbiota.
The minor pilin (PilV) is part of a shufflon that can exchange DNA cassettes at the 3’-end of the pilV gene. Eight different C-terminus variants of PilV can thus be generated by the action of a tyrosine recombinase.
In our most recent article, we showed that inactivating the major or minor pilins composing the T4P nearly abolishes the transfer of conjugative plasmid TP114 in the mouse gut microbiota. We also observed that there is more than one minor pilin...
