Want to base edit and no PAM in reach? Just create one! "Inception" is a one-shot event based on a canonical target site to create a de novo PAM and a 2nd editing upstream, relaxing steric dependencies. #CRISPR #baseedit with @kaisa_pakari, @WittbrodtLab

and furthered by comparison with other regenerating and non-regenerating fish like the surface and cave phenotypes of #astyanax. impressive talk at #GfE2024!

very nice talk from @Adelmann_L on #platynereis. She makes use of #scTranscriptomics over time during the course of regeneration and finds stem cell hotspots in different clusters 💪 #GfE2024

Great talk by Christof about one of the hallmarks of #DevBio. Brings back memories of the good time I spent on the organizer during my PhD dissecting its downstream left-right organizing activity. #xenopus #cilia #LROrganizer

In contrast to PAM-less editors, inception uses target-specific NGG-editors. Chaining 2 guides & 2 editors is predictive & efficient. Though simultaneously applied, mode of action is sequential. 2nd edit depends on the 1st - allowing editing 27-36nt upstream of the canonical PAM

ahhh, bench air! Dear #miniprep, how I missed you! You routine chance for self-optimization. You simple process that grants room to think while doing, that makes automated movements happen thanks to imprinted memory in my Medulla spinalis. Even after years of abstinence. Thank U!

© Thomas Thumberger (COS)
