People who don't follow cancer research often ask me why we haven't cured cancer. That perception masks a wonderful reality: We make amazing, stepwise progress every year, and the result is that many people live much longer today than they would have previously. Right now we're in the thick of the annual meeting of the American Society of Clinical Oncology, the biggest research meeting on new cancer medicines, and this morning a bunch of really important studies dropped. I'm going to review them here. This first image is the result for daraxonrasib, a treatment for pancreatic cancer that is generating consdirable excitement. The green line is the probability of living for patients who got the new drug; the gray one is the chemo control group. If you follow cancer drugs, a chart like this will make your breath hitch a little. I'm going to review these and some other data here.

What an interrupting standing #ovation looks like from the front of #ASCO26 when a revolution in science comes for #pancsm treatment #GIonc.

Cheers, chills, and a standing ovation when RASolute 302 showed unprecedented survival on daraxonrasib for patients with progressive pancreatic cancer Seldom do you sense you’re witnessing a historic moment in cancer care but this feels like ras targeting has arrived #ASCO26

As many other oncologists will also attest, we were taught this was a dead end It was, dogmatically, never going to work — kras was too much of a “greasy ball” to be targeted And yet here we are, with truly meaningful survival curves👇 Inspiration on multiple levels #ASCO26
What I love about this news is that this is not simply a new pancreatic cancer drug. It’s that KRAS, one of cancer biology’s most notorious undruggable targets, has finally become druggable. A powerful reminder that persistence in science can eventually unlock the impossible.

This is a moral issue for the oncology community. Were a drug or supportive care intervention to show OS curves like this, it would be SOC, and covered. For far less than costs of new drugs, our society could provide food for our patients. Perhaps our clinics need to do that.
Food insecurity is not just a social issue — it is a cancer outcomes issue‼️ Pts with MBC living in food deserts had distinct genomic profiles, including higher rates of RTK/RAS pathway alterations and CCNE1 copy number variants, along w/ worse survival outcomes — particularly among Black pts. A powerful reminder that biology and structural inequities are deeply intertwined. #HealthEquity #ASCO2026 @OncoAlert

Dr. Caicun Zhou at #ASCO26 with results from phase III OptiTROP-Lung05: first-line sacituzumab govitecan (sac-TMT, TROP2 ADC) + pembro vs pembro for PD-L1+ NSCLC (n=413). Sac-TMT superior with PFS NR vs 6.6m (HR 0.38) with benefit across subgroups.

It’s astonishing that CROWN 7y update shows mPFS and median time to IC PFS are still NOT REACHED: Surprisingly only 34% needed a DR and 5% TRAE>discontinuation. Current 4th gen trials will be criticised for inadequate control arms. #ASCO26 @ALKpositiveINT @BTOGORG

Impressive data on #nextgen #KRAS G12C inhibitor, Elisrasib, at #AACR26. ORR 73.5% with durable responses to >= 3y in treatment naive population! #LCSM @yonsei_u
Exciting data presented by @cbcbc1971 @yonsei_u on elisrasib (D3S-001), a next-gen KRAS G12C inhibitor in advanced #NSCLC: ➡️G12Ci-naïve: Observed response rate 73.5%, with durable responses seen out to 3+ years ➡️G12Ci-refractory (600mg QD): ORR ~31%, with 8 pts (25%) still on treatment ⚡ 70% of responders achieved first response by Week 6. Elisrasib received BTD & Fast Track Designation for #NSCLC by @US_FDA. A promising option in both treatment-naïve & post-G12Ci settings @KRASKickers #lcsm #AACR26

Dr. @riess_md at #AACR26 updates RMC-9805 (zoldonrasib) in KRAS G12D NSCLC. Very impressive. G12D is a particular need in lung cancer as many patients have no smoking history and do not fare as well with immunotherapy. Zoldonrasib is a mutant selective, RAS(ON) inhibitor.

#AACR26 Zoldonrasib in previously treated KRAS G12D NSCLC with RR 52%, DCR 93%, time to response 1.4m, mDOR not reached with median follow up of 13.1m, mPFS 11.1m, and 73% of pts with ctDNA clearance. Fantastic presentation, exciting drug for our patients!

👀Not all KRAS mutations behave the same. 🧬KRAS G12V shows a clear smoking-driven profile (94% smokers) with frequent TP53/STK11/KEAP1 co-mutations—yet still derives benefit from PD-1 blockade, particularly in PD-L1 high disease (rwOS ~30 months). Not the “IO-resistant” story we expected✅ @OncoAlert @OncoReporte @MedwatchKate @ManuelDomine @lung_connect @MedicalwatchHQ

People often ask how breakthroughs occur in cancer biology-often the story is more complex - the survival plot for myeloma outcomes is extraordinary - improvements come about in incremental steps - in my lifetime treatment of Myeloma has almost transformed into a curable disease

KRASG12C inhibition meets EGFR feedback Block both KROCUS trial. Fulzerasib + cetuximab in 1L NSCLC (phase 1b/2) in @TheLancetOncol ORR 69%, PFS 12.5 mo Rational combination. Promising early signal thelancet.com/journals/lanon… @OncoAlert
We thought KRAS was the engine So it was turned off But tumors adapt, rerouting the pathway In this preclinical study in KRASm NSCLC, loss of PTPRR activates EGFR, reactivating ERK and driving resistance to sotorasib/adagrasib. KRAS is blocked EGFR turns the pathway back on That’s why vertical blockade (KRAS + EGFR) may matter, remember that preclinical study 🇪🇸 ❓ Will panRAS inhibitors be enough? Preclinical data suggest probably not. Combos may be the answer aacrjournals.org/cancerresco… @OncoAlert @Aiims1742
