The best founders in the world like @elonmusk or @tobi “go ultra-hardcore on deletion and simplification.” Luca Ferrari says Bending Spoons employs this idea of “Relentless Simplification” across their businesses: “Humans have a tendency to add complexity and do things that destroy value.” “If you leave an organization, almost any environment unattended, and you don't provide guidance in this regard, it will tend to become more complicated. People will add parts. It could be expanding a team or adding a step to a process, or adding an entire new process. If it's a product, adding a feature to the product. New rules. It applies to almost any human endeavor.” “People tend to add pieces, they very rarely remove pieces.” “If you don't make a conscious effort to achieve simplicity, and avoid this increasing value-destroying complexity, that's how we got to our modern society with all the bureaucracy and complicated regulation.” “We have this principle whereby we ask everyone who works here, every time someone is suggesting that we should be adding complexity, the burden of proof is on those making that suggestion. The people who support the thesis that we shouldn't be adding complexity don't need to prove it. They just have to raise a flag and say, ‘I don't think we should.’ The burden of proof is on those who want to add complexity.” “The other part of relentless simplification is that we want people to be on the lookout for existing complexity and suggest that we should remove it.”

Moderna/Merck just ran a 1,137-patient Phase 3 trial where every single dose was unique to that patient's tumor. It worked. The pipeline: surgical resection → whole exome + RNA sequencing → ML neoantigen ranking → mRNA encoding up to 34 patient-specific targets → manufactured and shipped in 8 weeks. One drug, different sequence for every patient. The ML step is worth to note: the algorithm ingests WES + RNA-seq to identify somatic mutations, then predicts which of those will actually be immunogenic, ie, displayed on tumor cell surface and trigger a T-cell response. It's designed to keep learning from accumulated clinical and immunogenicity data across patients, not just per-patient. INTerpath-001 (Stage IIB-IV resected melanoma, 2:1 randomized): combination with pembrolizumab beat Keytruda alone on both primary (RFS) and key secondary (DMFS) at interim. Phase 2b at ASCO 2026 showed 49% reduction in recurrence/death, 59% in distant metastasis/death at 5 years. Phase 3 confirmed both. What this validates: — tumor-specific neoantigen prediction by ML works in a blinded trial at scale — 8-week personalized mRNA manufacturing is operationally real — effect is additive on PD-1 blockade, not redundant This is first positive Ph3 for individualized neoantigen therapy. First positive Ph3 for any mRNA cancer therapeutic. What a great time to live in!! This is the best time for AI & biotech!
Moderna and @Merck today announced positive topline results from the Phase 3 INTerpath-001 trial evaluating adjuvant treatment with intismeran autogene, a novel investigational mRNA-based individualized neoantigen therapy jointly developed by Merck and Moderna, in combination with KEYTRUDA® (pembrolizumab), Merck's anti-PD-1 therapy, in patients with completely resected Stage IIB-IV melanoma. PSA: If you are tired of Claude-lish or Chat-lish, tell your AI to read the Google Developer Docs Style Guide and build a skill that follows it It’s like magic. Suddenly your AI will make sense again! I’ve tried both and this is much better than the ASD-STE100 Simplified Technical English skill And friends at the labs: please invest more in AI comms skills! A brilliant agentic AI that cannot speak English is dramatically less useful and undermines the utility of RLVR developers.google.com/style

Been there done that. Great summary
Over the past 6 months we've maniacally prepped our repos to be coding agent friendly. Here are some things that worked. 1. Make CI blazing fast. Use every Rust, Zig, or Go ported tool that lets agents verify their work. This means UV, oxfmt, Typescript 7. Move integration tests to post merge hooks. 2. Trigger coding agents automatically based on triage labels. A coding agent should setup a proof of concept or repro steps automatically so an engineer can pick up the issue seamlessly. 3. Setup crons for things devs hate doing. Setup agents to fill SDK gaps, skill tuning, filling in critical regression checks. 4. Give agents ways to prove their work. Add screenshotting skills, agent-browswer, cloud storage for storing assets. 5. Make it possible to hermetically deploy your app, preferably multiple at a time. If a coding agent can deploy the app locally, the faster it can work. 6. Give the agents realistic production "simulation" data. Agents will work much better when they are working against data that looks like how your users use the product.

The great lie is that society is divided between rich and poor. The great truth, as David Friedberg puts it, is makers vs takers. Makers build, create, and deliver real value: houses, software, art, businesses, and everything that moves civilization forward. Takers watch, criticize, analyze, and politic. They push the lie that the rich hoard unfairly so the poor must seize it… all while positioning themselves to rule the chaos. As @friedberg tells his kids: “At the end of the day, if you made something and someone else valued it, you were a maker. That was an amazing achievement. That is a great day.” Takers thrive on division. Makers drive progress. Time to choose your side.

Today I woke up thinking about all the code I wrote early in my career, code that’s now nowhere or deprecated. It’s crazy how much time I spent picking variable names or writing good comments. I guess those of you using Claude can thank me.

.@nodejs has always been about I/O. Streams, buffers, sockets, files. But there's a gap that has bugged me for years: you can't virtualize the filesystem. You can't import a module that only exists in memory. You can't bundle assets into a Single Executable without patching half the standard library. That changes now 👇

The reason we have 12 hours cycles instead of 10 or 24 goes back ~4,000 years to the Babylonians, who used a base-60 number system. That’s why we also have 60 minutes and 60 seconds.🤯

lot of commercial OpenCode forks floating around that we get tagged on so gonna write up our positioning OpenCode is very intentionally MIT licensed - we hope to do such a good job that companies building coding agents opt to build on top instead of reinventing the undifferentiated parts it's tedious - getting things working across hundreds of LLM providers, dealing with quirks, collaborating with all the companies involved to improve things and then the platform work to get things working well on all environments thankfully we have a solid business model and can devote significant resources to doing all of this - including funding things like OpenTUI the forks that have popped up are confirmation we're starting to hit these goals so that's great - even when they're from competitors what's less great is they seem bent on passing off all this work as their own, even claiming they "built it from scratch" they've never communicated with us, never privately said "hey thanks for the work" every company can choose how they want to operate but this isn't very aligned with the spirit of open source and these small gestures of good will go a long way
