he spent years filing down cracked heels and one day noticed the cracks were gone
guy was about 90 days into daily GHK-Cu shots when he spotted it
his log:
- years of deep cracks on his heels and the balls of his feet
- used to grind and file the thick skin down because it kept coming back
- around day 90 of subq GHK-Cu he noticed the cracks were basically gone
- feet smoother and less dry too
- in the comments he gives it as 2 mg subq daily
- also credits it with fading his stretch marks and bringing back hair at his forehead
- says his skin is beyond glowing
- someone guessed a B vitamin or omega 3 gap · he says he was already on fish oil and B-complex with a lot of animal protein
- says the only recent change was "addition of peptides" · plural, and he never names the rest
- 18 years of smoking pot · calls himself a past and sporadic alcoholic · works out religiously
- asks a glutathione stacker if oral is easier · says his subq shots are massive and give him welts
what it is:
- GHK · glycyl-histidyl-lysine · a copper-binding tripeptide found in human plasma, saliva and urine
- Loren Pickart isolated it from human plasma in 1973 · it made old liver tissue build proteins like young tissue
- Pickart's 2015 review puts plasma levels around 200 ng/mL at age 20 and around 80 by 60
- same review lists faster wound healing in the foot pads of dogs · that's animal work
- mostly sold as a topical skin and hair ingredient
- not an FDA-approved drug by any route
- FDA had injectable GHK-Cu in compounding category 2 · immunogenicity risk, limited data in humans
- that injectable nomination was withdrawn in April 2026 · only non-injectable GHK-Cu went back to category 1, on 14 May 2026
- FDA says it will take GHK-Cu to its compounding advisory committee before the end of February 2027
so either a copper peptide finally fixed skin he'd been filing for years
or whatever else changed in those 90 days got credited to the one compound he named
the interesting part is the confound he wrote himself: "addition of peptides", plural, and the rest of the stack never shows up

nine years of stimulants and he still just laid there
guy asked for guanfacine instead of atomoxetine
r/NooTopics · FederalGovernmentUS · 9 Aug 2026
title says Intuniv · body says guanfacine 4 mg
his log:
- nine years on venlafaxine · Vyvanse, Ritalin, Concerta, dexamp rotating through
- stim arrived and he laid there · avoiding responsibilities and discomfort
- atomoxetine had a bad reputation in what he had heard
- asked for 4 mg guanfacine · left the other meds in place
- within months: masters program · more active than the year before · sleeping on time
- task initiation, his worst symptom, pretty much gone
- desk still becomes a mess · he cleans it after two days now · the pile used to sit because facing it overwhelmed him
- he reads that as unrecognized anxiety · calls guanfacine a game changer for that case
- post never says extended-release Intuniv vs immediate-release · silent on blood pressure, pulse, sedation
what it is:
- Intuniv · extended-release guanfacine · DailyMed label revised June 2025
- central alpha-2A agonist · about 15 to 20x affinity vs alpha-2B or 2C
- label: not a CNS stimulant · mechanism in ADHD not known
- same section: those central receptors cut sympathetic outflow · peripheral resistance and heart rate drop
- efficacy writeup on the label: ages 6 to 17 · alone and added to stimulants
- marketing start 2 Sep 2009 · Shire notice 9 Nov 2009 on EurekAlert gives the same FDA date and age range
- label header still says initial U.S. approval 1986 · that is the older immediate-release blood-pressure tablet
so either an alpha-2 add-on unstuck task initiation the stim stack never touched
or nine years of laying there finally got a story with a new pill in it
the interesting part is the confound: other medications stayed on, and the label's efficacy ages stop at 17

amino metabolite from the gym shelf claimed to kill his depression in one hour
guy ran agmatine sulfate for 9+ months
1 g morning · 1 g early evening
Not just an OP but also an OG worth checking out: sirsadalot · r/Nootropics (later founded NooTopics).
his log:
- hour 1 after first gram: stopped dwelling on negative thoughts · stopped lashing out · no longer wanted to die
- claims he could finally redirect thought patterns
- months in: less clingy / fear-driven · more goal-oriented · dropped daily masturbation addiction without ED
- claims learning and retention improved
- alcohol: every drink instantly felt like he skipped the dose · stopped drinking
- cites L-citrulline / L-arginine as possible blockers of the antidepressant effect
- zero claimed physical sides beyond NMDA-antagonist potentiation of other substances
- sometimes feels too relaxed · wonders if missing the old sadness range costs creativity
- not ready to stop to test that · plans to keep going until goals land
what it is:
- agmatine · arginine metabolite · sold as a gym / nitric-oxide adjacent powder
- NMDA / imidazoline / polyamine signaling lane in the literature · not an approved antidepressant
- the "what it is" is OP framing · not a clinical claim from us
- wrong-shelf mood drug energy: bodybuilding powder with an hour-1 mental flip in this log
so either a cheap amino metabolite hit a real NMDA mood switch in under an hour
or hour-1 miracle language is doing the heavy lifting over a nine-month clock
the interesting part is the kill switches he named: alcohol · maybe arginine cousins · and the creativity worry he will not test
n=1. grocery powder · hour-1 cure claim · nine months without coming off
people look at Mesocarb and read the left side first. a phenylisopropyl fragment sitting right there, unmistakable. the eye finishes the molecule before the brain does and files it next to amphetamine. that read is where the whole misunderstanding starts. the bottle said Sydnocarb, sometimes Sidnocarb. Soviet development lands in the early 1970s, anchored to GB patent # 1,262,830.
the fragment is real. the conclusion is wrong. the amine nitrogen is not hanging off a chain, it is inside the ring. middle of the molecule is a mesoionic sydnone imine with the charge pair delocalized across the five-membered ring, a structure you cannot draw neutral without breaking aromaticity. right side is phenylcarbamoyl. shape says rush, peak, comedown. pharmacology delivers a different curve.
the metabolism fight is live, for what it's worth. Melior's own patent filings state sydnocarb is metabolized to D-amphetamine in humans and animals, with the role of that metabolite in net in vivo effects unclear. the counterargument is that the prodrug attribution rode on older work using gas chromatography. pick your artifact.
Gruner's group published the wake and release work in JPET in 2011. amphetamine emptied dopamine from rat synaptosomes at an EC50 of 0.10 micromolar. sydnocarb did not, up through 100 micromolar. human DAT binding Ki was 8.3 nanomolar in that assay. human NET sat at 10.1 micromolar, about twelve hundredfold weaker. no binding to 64 other targets at 10 micromolar. sleep EEG in rats showed 10 mg/kg sydnocarb and 1 mg/kg amphetamine raising wakefulness comparably, with no amphetamine-like rebound hypersomnolence up to 30 mg/kg. theta power rose. amphetamine raised it too, so the differentiator is the missing crash, not the theta. the authors framed theta as a cognitive electrophysiology correlate. it is their framing. it is not a human smart-drug proof.
Aggarwal's 2021 Biomedicines paper moved the story onto allostery. sydnocarb binds an extracellular site A2 on human DAT, the same vestibule pocket as their modulator KM822, outside cocaine's orthosteric plug. cocaine failed to protect the W84C mutant. sydnocarb did. uptake IC50s landed at 0.49 micromolar at DAT, 34.9 at NET, 494.9 at SERT. seventyfold and thousandfold. Vmax dropped, Km held. noncompetitive. and it degraded the other two: cocaine's IC50 moved from 0.177 to 8.40 micromolar under 2 micromolar sydnocarb, amphetamine's from 0.166 to 2.60. negative allosteric toward both.
microdialysis fills the shape. Gainetdinov's 1997 striatal work put sydnocarb at a modest dose-dependent rise, up to about 350 percent of control, holding for six hours. amphetamine spikes higher and faster, then drops. at equimolar doses sydnocarb gave less stereotypy and less hydroxyl-radical signal. but the same paper found the efflux tetrodotoxin-sensitive and calcium-dependent, which reads as facilitated vesicular release, not a clean uptake block. and Gruner's 8.3 nanomolar binding Ki sits sixty-fold from Aggarwal's 0.49 micromolar uptake IC50 across different assays and cell lines. the clamp story is the best available story. it is not a closed one.
Soviet clinics mapped soft edges of that curve under period labels: asthenia, apathy, adynamia. historical indications. Western approvals never followed. US Schedule I, UK Class C, WADA prohibited list. Russian production wound down in the 2000s. the public record does not pin a clean safety kill against a quiet business exit, so the motive stays soft.
Melior picked the compound back up for Parkinson disease. the lead claim was L-DOPA-induced dyskinesia. the 2015 US patent covers attenuating dyskinesia while improving levodopa's therapeutic benefit, and Adhera's planned Phase 2a was designed to enroll PD patients with LID. movement plus non-movement aspects, sleep and daytime sleepiness included, was the wider sponsor framing sitting on top of that. the Michael J. Fox Foundation awarded Melior Discovery a PD repositioning grant in August 2011, under MJFF's first repositioning-specific RFA, six awards totaling $2.4 million. codes became MLR-1017 for the racemate and MLR-1019 / armesocarb for the active enantiomer. Adhera licensed armesocarb on July 28, 2021 and announced API manufacturing aimed at the Phase 2a. Melior terminated that license effective January 12, 2024 in an SEC 8-K, citing Adhera's failure to continue commercially reasonable efforts. clinical-stage reuse with an interrupted partner path. unfinished efficacy read. no approved Parkinson drug.
wake and excessive daytime sleepiness ride the same slow dopamine curve. the JPET sleep work showed more wake in the window without amphetamine's crash sleep. that is why sponsor language keeps reaching for wake and EDS in Parkinson. if the allosteric DAT story holds, the cognitive use-case is vigilance and effort under load. Soviet "nootropic" labeling was brochure language sitting on a DAT mechanism Western pharmacology only sharpened recently.
the ring itself is having a moment, incidentally. IRBM in Pomezia published the first orally efficacious allosteric flavivirus protease inhibitor in Nature Communications this February. the series carries an N-acylsydnone imine, binds a previously undescribed allosteric pocket on Zika NS2B-NS3, and locks the enzyme catalytically dead. same weird zwitterionic heterocycle, completely unrelated target, allosteric again. structures are in the PDB.
cursedchemistry boards float Nitromemantine next to Mesocarb: memantine's adamantane cage carrying an appended nitrate group out to the receptor's extracellular redox thiols, channel block coupled to allosteric redox modulation (Takahashi et al., Scientific Reports 2015, focal cerebrovascular model). the memantine nitrates were less potent than memantine at the receptor and still outperformed it in the stroke model. different molecule. different plot. rare mechanisms travel in pairs on chemistry boards long before they travel in clinics.
the cursed part is the date. Erdö, Kiss and Rosdy published dopamine uptake inhibition by mesocarb in 1981, in the Polish Journal of Pharmacology and Pharmacy. the signal was in print for forty years while English stimulant talk stayed stuck on release versus reuptake orthodoxy.
