🥳🥳🥳 Our study on engineering enzymes to break C–F bonds is now out in @angew_chem! Congrats to the brilliant & dedicated Suzanne Jansen for taking this first step toward the future bioremediation of #PFAS & other contaminating organofluorides!

🚨New preprint! What a difference one C-F bond can make!🚨
Fluoroacetate dehalogenases (FAcDs) are remarkably good at breaking C-F bonds. But add just ONE more C-F bond to the substrate and suddenly the enzyme is inhibited.
So what’s going on? (1/4)

Substituting this histidine with a non-canonical histidine analog or Gln can overcome the inhibition! A beautiful example of modern mechanistic enzymology and an important step toward our longer-term goal of making FAcDs fit for PFAS degradation. 🌍
One C-F bond at a time! (3/4)
Huge credit to @suzanne__jansen, who led this work. But without Luis Guerra & Manuel Müller from @lab_mueller, as well as Cristina Duran & @silviaosu, we could have never provided experimental and computational evidence for isoaspartate formation! 🤝 (4/4)
🧵 New review out in Chemistry – A European Journal!

With #PFAS and other organofluorides wreaking havoc, we set out to engineer natural defluorinases for man-made organofluorides. All credit goes to the amazing @suzanne__jansen, who has outdone herself in this project! (1/5)

Forcing populations of bacteria expressing tens of thousands of distinct FAcD-variants to utilize various organofluorides as sole carbon source, we elicited a panel of FAcD variants with improved activities and drastically altered substrate profiles. (4/5)

So glad to see this out! By giving a strong fluorescence signal to read out NAD(P)H concentration, SAFRAN will greatly extend the range of reactivities comfortably assayable in droplets. This will make droplet microfluidics a much more versatile tool for biocatalyst engineering!

Two weeks ago, we had a great group outing, including a city tour through Utrecht, a Mario Kart Wii tournament, and some Rock Band Wii fun! I am truly privileged to work with such a group of talented, dedicated, and smart individuals.
