$CGON BofA frame PIVOT-006 as asymmetric, with a HR closer to 0.45-0.55implying ~20% upside to base case and a HR >0.7, most likely on a stronger-than-expected control arm, implying ~30% downside, and a clean readout followed by solid 12-month data would, in our view, cement creto as a backbone therapy across the NMIBC spectrum heading into 2027.

Their manufacturing processes are also fundamentally different.
$OBX uses potent T-cell stimulatory agents (4-1BB agonist, OKT3, IL-21, etc.) during manufacturing, resulting in a predominantly CD8+ T-cell product. In contrast, $IOVA Amtagvi exhibits a much broader range of CD4/CD8 ratios.
This could have significant implications for response kinetics, durability, and indication-specific efficacy.
I think this is an overlooked aspect of the $OBX story. Everyone seems focused on mbIL-15, reduced lymphodepletion, IL-2 elimination, biopsy requirements, etc., while paying relatively little attention to the actual cellular composition of the final product.
OBX's manufacturing process appears to substantially deplete CD4+ T cells, which may account for ~20% of tumor-reactive T cells in melanoma but potentially ~50% in NSCLC.
That difference could be particularly important given the role of CD4+ T cells in supporting durable antitumor immunity and T-cell memory formation.
Of course, not all CD4+ T cells are beneficial—some are immunosuppressive Tregs. But broadly eliminating CD4+ cells could mean losing important antitumor functions along with the suppressive ones.

Weekly recap — what X wouldn't shut up about.
Week of Nov 2
1️⃣ $MRNA
Fri: Moderna's third-quarter report. It made a profit this quarter and raised its full-year sales outlook.
💊 Pharmacist note: its new flu shot, mFLUSIVA, was approved in August, after most flu-shot orders were already placed, so this season's U.S. launch is limited.
2️⃣ $NVO
Wed: Novo Nordisk raised its full-year outlook on stronger sales of Wegovy, Ozempic and its other GLP-1 drugs.
💊 Pharmacist note: the Wegovy pill is now sold in 8 markets. A pill skips the needle, not the stomach side effects, so slow dose increases still matter.
3️⃣ $PFE
Tue: Pfizer's Q3 sales were $16.6B, flat with last year, and it raised its 2026 forecast.
💊 Pharmacist note: COVID products shrank. Eliquis, the blood thinner, was one of the drugs that grew.
Dates first. Hype later.
Tune in Sunday for the weekly Issue.

You don’t short a company when a CEO comes out like this and buried in the last sentence of the press release is a government acronym that says the US government is also done waiting and is going to push this company through

$IOVA Barclays just raised its price target from $18 to $19 while maintaining its Overweight rating.
That’s roughly 145% upside from yesterday’s $7.74 close.
Citizens also raised its target today from $8 to $12.
Two firms raising targets following Iovance’s latest earnings. Wall Street clearly isn’t unanimous on valuation, but the bullish analysts continue to see substantial upside. 👇👇
