Chemical #probes are important tools for validating drug targets. Covalent inhibitors & molecular degraders are expanding the druggable space. We now propose quality criteria for covalent & degrader probes: @ACSBioMed pubs.acs.org/doi/pdf/10.1021… @Chemical_probes @thesgconline #TPD

Finally I read up on #DCAF1 based #PROTACs @NovartisScience. A recently disclosed non-covalent WD40-binding DCAF1 ligand enabled degradation of BRD9, Csk, Limk2, TEC, TNK2 & BTK. Hijacking essential DCAF1 may break resistance to CRBN PROTACs. @BioRxiv #TPD biorxiv.org/content/10.1101/…

Interesting paper by the @london_lab in @J_A_C_S. They developed sulfamate acetamides as interesting new cysteine-directed electrophiles that allow development of covalent inhibitors as well as applications in covalent ligand-directed release (CoLDR). doi.org/10.1021/jacs.2c08853

2. Sulfamates can be used for site-specific traceless labeling of proteins. We demonstrated labeling BTK C481 with modifications as small as a single methyl or propargyl. The labeling is concomitant with the elimination of the inhibitor, and the protein stays active. 10/n

Special credit first - This is the final chapter in the CoLDR trilogy by @reddi78 who is the lead and co-corresponding author (pubs.acs.org/doi/10.1021/jac… ; pubs.acs.org/doi/10.1021/jac… ). A fantastic and original chemist who is now on the job market! Go get him! 2/n

Paper alert 🚨 We report @J_A_C_S sulfamate acetamides as a new electrophilic “warhead” for late-stage optimization of covalent inhibitors and for covalent probes suitable for cells and in vivo administration. pubs.acs.org/doi/10.1021/jac…
