We show that H3 ubiquitylation, its subsequent acetylation on lysine 56 by the histone acetyltransferase Rtt109, and its deposition on newly synthesized chromatin by the histone chaperone Asf1 are crucial to prevent toxicity of such seDSBs, which are resolved by HDR.
