What if a simple blood test could help us decide when to switch treatments in advanced melanoma? 🧬 Join OncoAlert & our Faculty Dr. Mario Balsa🇪🇸 for this #WeekinOncology #Melanoma as he discusses how the CAcTUS🌵 trial explores how circulating tumour DNA (ctDNA) could help personalise treatment sequencing for patients with BRAF-mutant melanoma—potentially allowing clinicians to make faster, data-driven decisions. Use of circulating tumour DNA to prospectively guide a switch from targeted to immune therapy in BRAF mutant advanced melanoma: the randomised phase II CAcTUS trial The CAcTUS phase II trial explored whether circulating tumour DNA (ctDNA) could guide treatment sequencing in patients with BRAF-mutant advanced melanoma. Patients received targeted therapy (TT) or checkpoint inhibitor immunotherapy (CPI), with treatment switching based on progression or an 80% reduction in BRAF variant allele frequency. ctDNA results were delivered rapidly, successfully guiding treatment decisions. Findings suggest ctDNA may identify treatment response early and help optimise sequencing, with no new safety concerns observed. buff.ly/x6xm9Mh @BeckiLee

Key finding #3: longitudinal ctDNA monitoring provided early signals of both treatment response and resistance, often before radiological changes were seen. 📈

5/5 This was a small study and not powered to determine efficacy, but it provides a foundation for future ctDNA studies in melanoma and treatment sequencing approaches. Thanks to funders, participating sites, and especially the patients involved.

It's day 17 of our 2025 #AdventCalendar!! 🎅 At the start of the year, we saw the first patients recruited to the @CR_UK-funded DETECTION-2 trial, seeing if a simple blood test can spot #melanoma coming back, even if a scan looks normal. Find out more - bit.ly/3Dfc8ty

We have the cover of Cell today. Craters on the surface of melanoma serve as an immune hub for CD8+ T cells. A timely highlight as we roll into Thanksgiving.

Where do cancers start? In the liver, not all cells are the same. The liver is divided into 3 “zones,” and each specializes in different metabolic tasks. We show in @Science that where a mutation occurs in a liver lobule can determine a cell’s fate. science.org/doi/10.1126/scie… (1/9)

Finally, someone has solved a real problem with AI! No more having to take a paper in the format for a journal that rejected you, and reformat it for a new journal. Well done!! formatmypaper.com/
