#ClonalHematopoiesis of indeterminate potential (CHIP) is common in older individuals, but its connection with aortic valve stenosis was unclear.
Here, @AlexBickMDPhD @StefanieDimmel1 @AndreasZeiher @cpi_exstra reveal an association between TET2 mutations and aortic valve stenosis (n=886,378), showing #CHIP activates procalcific pathways in macrophages and promotes aortic valve stenosis: jci.org/articles/view/171634

CHIP (TET2 and ASXL1 mutations) heightens risk of incident aortic stenosis through oncostatin M (OSM) signaling. OSM is in the family of IL-6 cytokines and signals through gp130 - makes me wonder what the effect of the IL-6R hypofunction variant/IL-6 blockade may be.
We still don't measure CHIP in the clinic (blood stem cell clones) which occur frequently with age and are a risk factor for cancer and heart disease. Here's how CHIP can lead to a calcified aortic valve. @jclinicalinvest
