🎉Check out our largest CHIP GWAS to date of >1M participants, shedding light on how germline variation governs which somatic clones in blood expand with aging🧬 In parallel, we conducted CHIP PheWAS, linking CHIP to hematologic, cardiovascular, infectious, and other conditions🩸🫀🦠 medrxiv.org/content/10.1101/… @M_Mesbah_Uddin @AlexBickMDPhD @pnatarajanmd
Our preprint led by @M_Mesbah_Uddin @ZhiYu_ACGT and with @AlexBickMDPhD describes the germline genetic drivers and phenotypic associations of clonal hematopoiesis of indeterminate potential across >1M individuals medrxiv.org/content/10.1101/… @medrxivpreprint

Very excited to share our study spearheaded by A. Misra & @AniruddhPatelMD describing the development & validation of our clinical multi-CVD polygenic risk score test, including how it might inform CVD prevention sciencedirect.com/science/ar… @JACCJournals Any clinician can order this @MassGenBrigham @broadinstitute test today: massgeneralbrigham.org/en/re… We used an integrated framework we previously developed and described to maximize performance and score stability, and allow for versioning as genomic interpretation continually improves. cell.com/cell-genomics/fullt… nature.com/articles/s41467-0… The score was optimized in the nationwide @NIH @AllofUsResearch Program to enhance generalizability across communities, with validation in the @MassGenBrigham Biobank @amandajowell @buutrg @aklfahed @shaan_khurshid @patrick_ellinor @nialljlennon @HornsbyWhitney @EWkarlson

Exciting research news! DoM fellow, Dr. Michael Raddatz, and his team published groundbreaking research in @JClinicalInvest uncovering how CHIP mutations increase the risk of aortic valve disease. Their work could lead to new preventive treatments: uclahs.fyi/49JfFhi

Happy to see @jclinicalinvest highlighting our work on CHIP and aortic valve stenosis. Short thread below ⬇️ xitter.cf/MichaelR21/status/1990…
#ClonalHematopoiesis of indeterminate potential (CHIP) is common in older individuals, but its connection with aortic valve stenosis was unclear. Here, @AlexBickMDPhD @StefanieDimmel1 @AndreasZeiher @cpi_exstra reveal an association between TET2 mutations and aortic valve stenosis (n=886,378), showing #CHIP activates procalcific pathways in macrophages and promotes aortic valve stenosis: jci.org/articles/view/171634 The figure shows increased aortic valve calcification (OSM, green) and inflammation (S100A9, red) in a CHIP TET2-mutant mouse model; nuclei (DAPI, blue).

CHIP (TET2 and ASXL1 mutations) heightens risk of incident aortic stenosis through oncostatin M (OSM) signaling. OSM is in the family of IL-6 cytokines and signals through gp130 - makes me wonder what the effect of the IL-6R hypofunction variant/IL-6 blockade may be.
We still don't measure CHIP in the clinic (blood stem cell clones) which occur frequently with age and are a risk factor for cancer and heart disease. Here's how CHIP can lead to a calcified aortic valve. @jclinicalinvest jci.org/articles/view/171634
