I sat down with Nautilus $NAUT CEO Sujal Patel and CFO Anna Mowry Sujal sold Isilon to EMC for $2.25B. Now he wants to do for the proteome what sequencing did for the genome Platform, launch, cash runway, and proteomics as the missing layer of “biology is code”👇

We’re finally able to see tau at the single-molecule level and more deeply explore neurodegenerative drivers. Thanks for the collaboration, @NautilusBio! There is real patient impact potential in this work. Read the paper here: nature.com/articles/s41592-0…
We are incredibly proud to announce that our paper demonstrating the large-scale single-molecule analysis of tau proteoforms by Iterative Mapping has been published in Nature Methods! 📰 Press release: globenewswire.com/news-relea… 📘 Paper: nature.com/articles/s41592-0… This paper shows that single-molecule proteomic analysis reveals entirely new facets of biology. Specifically: 🔵 Iterative Mapping enables accessible and scalable proteoform analysis that is not possible on other platforms — Proteoforms are the many distinct molecular forms of proteins that exist in cells. Many can be derived from a single protein and may have vastly different functions depending on their combinations of isoforms, sequence variations, and post-translational modifications. 🔵 Iterative Mapping precisely, sensitively, and accurately quantifies tau proteoforms over a wide dynamic range with high reproducibility (≤5.5% median CV). 🔵 Model systems commonly used in neurodegenerative disease research have distinct tau proteoform profiles. 🔵 In a small cohort, tau proteoform analysis differentiates samples donated by cognitively normal individuals from those donated by people with Alzheimer’s disease and related dementias. 🔵 Preliminary evidence suggests there are ordered pathways underlying the creation of tau proteoforms (they are not created randomly). This implies researchers may be able to prevent the generation of pathological tau proteoforms by disrupting these pathways! Congratulations to everyone at Nautilus and beyond who made this groundbreaking work possible. We’d particularly like to recognize our amazing collaborators including: Researchers from the Regenerative Research Foundation/Neural Stem Cell Institute: Sally Temple, Taylor Bertucci, David Butler, and Steven Lotz Researchers from The Ronald Loeb Center for Alzheimer’s Disease Research Icahn School of Medicine at Mount Sinai: Joel W. Blanchard, Ana Forton-Juarez, Emily Sartori, and Jessica Schwarz Researchers from Genentech: David Arnott, Joanna Lipka, Nikhil Pandya, Lionel Rougé, TJ Wendroff, Don Kirkpatrick, and Alexis Rohou Your expertise, deep understanding of tau and neurodegenerative disease, and innovative spirits made this work a powerful demonstration of the power of Iterative Mapping.

Had a great conversation with @paragmallick about the #proteoform . Short version: we need to sequence the human #proteome, like we did the #genome 20 years ago! #Proteomics #Biotech #Science
The active forms of proteins found in biological systems aren't simple strings of amino acids, but are the single-molecule products of many processes including splicing, phosphorylation, ubiquitination, and more. These "proteoforms" drive biology and thinking about them leads to the intriguing question, "Are proteins a myth?" (No, but we certainly need to know more about them!) Nautilus Co-Founder and Chief Scientist @paragmallick and Northwestern University Professor, proteomics expert, and proteoform pioneer @NLKProteomics discuss this question and many more aspects of proteoform biology on the latest episode of the Translating Proteomics podcast. Join the conversation and find real evidence for the incredible importance of proteoforms here: nautilus.bio/blog/translatin… #Proteoforms #Proteoform #Proteomics #Proteome #Biotech #Biomarkers #DrugDevelopment #Biology‌‌‌‌‌ #Podcast #Science
