My thoughts on the phase 3 results of Alzheimer's drug lecanemab from Biogen and Eisai, presented at #CTAD #CTAD22 and published this evening in the @NEJM. Major takeaway – this is NOT the breakthrough we have been waiting for. Key points:

New work from our lab shows how pericyte loss during brain aging contributes to blood flow disruption and capillary rarefaction. A repair strategy to mitigate these effects exists (pericyte remodeling), but needs help in the aged brain. biorxiv.org/content/10.1101/…

Considering the extent of pericyte loss seen in Alzheimer's disease and related dementias, we need to find ways to protect these cells early in disease progression.

my mental health is not loving the constant oscillation between “this is such a great idea - i’m a genius” and “this is complete garbage and will never work” while writing a research proposal lol

When I started my PhD, every other student in the lab was frustrated and burnt out, and I naively believed that my time would be different, that these were individual problems and I would somehow be a 'better,' more successful student. Oh how wrong I was...

What a great way to start the day, with a meeting with our sorting team!! We sort 600+ abstracts every month into categories within the field of #alzheimer's research. These categories make our episodes, accessible wherever you listen to #podcasts! #scicomm

This table has to be the most damning and depressing reflection of both the amyloid hypothesis and the current front-runners for Alzheimer’s disease modification that I can think of. By any objective mood measurement scale, my rating drops by 30% when I look at this.
The anti-amyloid pipeline has been vibrant for 2 decades. Amyloid lowering was achieved in ~75% of 40 trials. Yet in ~40%, outcomes favored placebo over treatment. We have been unable to falsify the amyloid hypothesis because of hope, belief, and, yes, greed.

My journey thus far… High school: What is Alzheimer’s? Undergrad: What is there than we can do to help find the cure for Alzheimer’s? PhD 2020: What is there we can do to find Alzheimer’s? PhD 2021: What is Alzheimer’s? PhD 2022: What?

It is out in the @officialEANM journal! What's the biological interpretation of FDG-PET brain hypometabolism? In this work led by @andreia_rocha_ we mimicked dementia-like cortical FDG hypometabolism by only reducing glutamate transport in astrocytes.bit.ly/3rrfrW2 1/+

A large multinational meta-analysis says that a third of cognitively normal people over 70 walk around with amyloid plaques in their brains. ow.ly/lMey50HNaye
PhD students tend to choose a lab based on the topic, but a creative and supportive environment is more important. If everyone in the lab is miserable, you will probably be miserable there too; and if they are happy and thriving, you'll also likely feel good about yourself there.
